same illusion of continuous motion as we have when watching movies, which are also
composed of separate images.
When we click to record an image, we say that the image is grabbed or acquired. Unlike
the live image, the grabbed image is saved in more permanent memory of the computer
for later use in image analysis and measurements.
We can read in the instructions for some comet software packages that measurement is
possible on live or imported (previously recorded) images. Measurement of a truly live
image is not possible, because such an image does not exist in digital technology. The
computer can only perform a measurement on a grabbed image, i.e. a saved image file
where all grey values for all pixels are strictly defined numbers. There are, however,
essentially two approaches to measurements. We can have a slide on the microscope, find a
comet, record an image and measure it immediately (live mode). Or, we can record images
of comets, save them in the computer memory, on a hard disk or on a CD, and do the
measurements later. If we have the possibility to do “live” measurements, our camera is
integrated in the software so that the computer can communicate with it, and we can both
record and measure images without exiting the program. Therefore, if we want to do
“live” measurements, we have to make sure when buying the equipment for the comet
assay that the software package supports our camera.
